Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by motor and non-motor impairments. Mild cognitive impairment (MCI) occurs in approximately half of PD patients and progresses to dementia. Currently, there are no effective treatments to halt or reverse cognitive decline in PD. In the present study, we investigated whether the novel antidepressant and antianxiety drug, venlafaxine, a serotonin-norepinephrine reuptake inhibitor (SNRI), could be neuroprotective in an animal model of PD. Male C57BL/6 mice were treated with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) for four consecutive days, and subsequently were treated with either vehicle or venlafaxine (20 mg/kg/day) for three weeks. The results indicate that treatment with venlafaxine significantly attenuated motor deficits in the MPTP-treated mice and rescued dopaminergic neurons from apoptotic death. This improvement in motor performance was associated with a significant increase in the activation of the Nrf2 antioxidant signaling pathway and a suppression of the JNK and p38MAPK stress-activated signaling pathways in the prefrontal cortex of the mice. However, the antidepressant effect of venlafaxine was not apparent.
